Resumo(s)O recurso às plantas aromáticas e medicinais (PAM) é uma estratégia milenar usada para tratamento, cura e prevenção de doenças. Algumas PAM, como resultado do seu metabolismo secundário, produzem óleos essenciais (OE), que dadas as suas propriedades, são utilizados cada vez mais na indústria farmacêutica. A sua capacidade antimicrobiana mostra-se como uma solução promissora no combate a bactérias, principalmente aquelas em que o uso indiscriminado de antibióticos provocou resistência à terapêutica, bem como, no combate a doenças infeciosas virais e fúngicas. Com o presente trabalho pretendeu-se conhecer a atividade antimicrobiana dos OE das plantas Poiretia bahiana, Acritopappus confertus e Cuphea carthagenensis oriundas da Chapada Diamantina na Baía, Brasil. Numa primeira fase testaram-se os referidos OE, em concentrações que variaram de 1000 a 4000 ppm, sobre a levedura Saccharomyces cerevisiae e as bactérias Escherichia coli e Bacillus subtilis, em ensaios de difusão em meio sólido e ensaios em meio líquido. Verificou-se que o OE de P. bahiana tem um efeito fungistático e fungicida sobre S. cerevisiae e um efeito bacteriostático sobre E. coli e B. subtilis. O OE de A. confertus apenas tem efeito bacteriostático sobre B. subtilis. O OE de C. carthagenensis não tem qualquer efeito sobre os microrganismos testados. Foi identificada a levedura S. cerevisiae como o microrganismo mais sensível aos OE testados e o OE de P. bahiana como o mais ativo. Na segunda fase do trabalho testou-se o OE de P. bahiana em mutantes deficientes em genes que codificam enzimas envolvidas na defesa antioxidante, SOD1, SOD2, CTT1 e em genes que codificam proteínas envolvidas na morte celular apoptótica, PEP4, AAC1,2,3, YCA1 e AIF1, de S. cerevisiae. Foi ainda testado um mutante deficiente respiratório, Rho0. Os resultados mostraram que OE induziu a produção de ROS na levedura S. cerevisiae e, embora, a mitocôndria não aparente ser o principal local de produção, terá um papel preponderante na capacidade da célula remover estes radicais. Os resultados sugerem ainda que a enzima Sod1 é essencial para proteger a célula do stress oxidativo promovido pelo OE, enquanto a Sod2p e a Ctt1p terão um papel menos relevante. OE de P. bahiana induziu um processo de morte celular programada, aparentemente do tipo apoptótico, mediado pelas proteínas Aac1,2,3 e Aif1, que têm um papel relevante no processo. Ao invés, as proteínas Pep4 e Yca1 não parecem ter um papel importante no processo de morte induzido por este OE.
Resorting to aromatic and medicinal plants (AMP) is a millenary strategy used to treat cure and prevent diseases. Some AMP, as a result of their secondary metabolism, produce essential oils (EO), that, due to their properties are being increasingly used in the pharmaceutical industry. Their antimicrobial activity appears as a promising solution in combating bacteria, especially those on which the indiscriminate use of antibiotics caused therapeutical resistance, as well as in the fight against viral and fungal infectious diseases. The present work aimed at evaluating the antimicrobial activity of EO of Poiretia bahiana, Acritopappus confertus and Cuphea carthagenensis from Chapada Diamantina in Bahia, Brazil. In a first stage, these EO were tested in concentrations that varied from 1000 to 4000 ppm, on Saccharomyces cerevisiae yeast, on the gram negative bacteria Escherichia coli and on the gram positive bacteria Bacillus subtilis, on diffusion assays in a solid medium and in a liquid culture medium. It was observed that EO of P. bahiana has a fungistatic and a fungicidal effect on S. cerevisiae and a bacteriostatic effect on E. coli and B. subtilis. The EO of A. confertus only has bacteriostatic effect on B. subtilis. The EO of C. carthagenensis has no effect on tested microorganisms. The yeast S. cerevisiae was identified as the most sensitive microorganism to the tested EO and the P. bahiana as the most active OE. In a second stage of the study we tested the P. bahiana EO on S. cerevisiae mutants deficient in genes that code for enzymes involved on antioxidant defense, SOD1, SOD2, CTT1 and on genes that code for proteins involved in apoptosis, PEP4, AAC1/2/3, YCA1 e AIF1. A respiratory deficient mutant Rho0 was also tested. The results showed that the EO induced the production of ROS in the S. cerevisiae yeast and, although, the mitochondria doesn´t appear to be main location of ROS production, it has a determinant role on the cell´s capacity to remove these radicals. The results further suggest that Sod1 enzyme is essential to protect the cell from oxidative stress caused by EO, while the Sod2p and the Ctt1p have a less relevant role. EO of P. bahiana induced a programmed cell death process, apparently apoptotic, mediated by Aac1/2/3 and Aif1 proteins that have a relevant role in the process. On the contrary, the Pep4 and Yca1 proteins appear not to have an important role in the death process induced by this EO.
