Resumo(s)Lactoferrin (Lf) is an iron-binding glycoprotein with multiple biological functions, including an anti-carcinogenic activity. It has been shown that Lf triggers apoptosis in both yeast and mammalian cell lines. Although this ability may explain Lf anti-tumoral activity, the underlying molecular mechanism still remains obscure. Vacuoles, like lysosomes in mammalian cells, are the major cellular degradation systems of the yeast cell. Their acidic lumen is generated and maintained by the vacuolar/lysosomal membrane V-H+-ATPase, which is responsible for pumping H+ against electrochemical gradient into the lumen at the expense of ATP. These organelles also participate in signaling pathways to apoptosis, which has instigated the search for drugs targeting these compartments ultimately aiming to overcome apoptosis evasion, one of the hallmarks of cancer cells. Therefore, whether or not bovine lactoferrin (bLf) could act directly on the yeast vacuole without the involvement of upstream effectors deserves to be investigated. This could provide further hints on the effect of bLf on lysosomes. To this end our studies were focused on isolated vacuoles from the yeast Saccharomyces cerevisiae and isolated lysosomes from rat liver. Several structural/physiological parameters were measured in response to bLf by flow cytometry and spectrofluorimetry with specific fluorescent probes, including transmembrane H+ gradient and lysosomal/vacuolar membrane permeability. We also investigated the effect of bLf in V-H+-ATPase pumping activity in yeast vacuole vesicles. Later we extended these study to isolated lysosomes and also assed the effect on the VH +-ATPase hydrolytic activity. The effect of acetic acid on the vacuolar function was also addressed in the present study following data from our group showing that Pep4p protease is released from the vacuole into the cytosol in cells undergoing acetic acid induced apoptosis Flow cytometric analysis of intact S. cerevisiae cells stained with the fluorescent probe carboxy-DCFDA were performed to further characterize the partial permeabilization of the vacuolar membrane induced by acetic acid.
A lactoferrina (Lf) é uma glicoproteína ligante de ferro com múltiplas funções biológicas, incluindo atividade anticancerígena. Tem sido descrito que a Lf desencadeia a apoptose tanto em células de levedura como em linhas celulares de mamíferos. Apesar desta capacidade poder explicar a atividade anticancerígena da Lf, os mecanismos moleculares subjacentes permanecem obscuros. O vacúolo da levedura, assim como os lisossomas das células de mamífero, constituem o principal sistema de degradação intracelular. O carácter acídico do lúmen destes organelos é conseguido pelo bombeamento de H+ contra o gradiente electroquímico mediado pela V-H+-ATPase à custa do consumo de ATP. Os vacúolos e os lisossomas participam ainda em vias de sinalização da apoptose, o que tem instigado a procura de drogas direcionadas para estes compartimentos celulares, com o objetivo final de superar a evasão das células cancerígenas à apoptose. Assim, pretende-se investigar se a lactoferrina bovina (bLf) pode atuar diretamente no vacúolo, sem o envolvimento de efectores a montante. Os resultados poderão fornecer pistas sobre o efeito da bLf nos lisossomas. Para este fim, vacúolos purificados de Saccharomyces cerevisiae e lisossomas de fígado de rato foram os alvos de estudo. Vários parâmetros estruturais e fisiológicos foram medidos em resposta à bLf por citometria de fluxo e espectrofluorimetria com sondas fluorescentes específicas, incluindo o gradiente de H+ transmembranar e a permeabilização da membrana vacuolar. Foi estudado também o efeito da bLf na atividade de bombeamento da V-H+- ATPase em vesículas de vacúolo de levedura. Posteriormente avaliou-se o efeito da bLf na atividade hidrolítica da V-H+-ATPase de lisossomas isolados. O efeito do ácido acético na função do vacúolo foi também estudado no seguimento dos resultados obtidos pelo nosso grupo de investigação que mostraram que a protease Pep4p é libertada do vacúolo para o citosol em células que sofrem apoptose induzida por ácido acético. Os estudos foram efetuados com células inteiras de S. cerevisiae por citometria de fluxo com a sonda fluorescente carboxy-DCFDA para caracterizar a permeabilização parcial da membrana vacuolar induzida pelo ácido acético.
