Resumo(s)A utilização da tecnologia MEMS no fabrico de sondas capazes de interagir com o sistema nervoso ao nível celular através de estimulação ótica é já uma realidade. Porém, estes microssistemas possuem limitações a diferentes níveis. As sondas neuronais dotadas de estimulação ótica (denominadas de “optrodes”) através de μLEDs possibilitam resolver o problema da liberdade de movimentos do sujeito em estudo, pois permitem o acoplamento de um sistema de comunicação sem fios. O problema que o tema desta dissertação visa resolver, é monitorizar o aquecimento colateral do tecido neuronal devido à utilização de μLEDs em optrodes para estimulação cerebral profunda. As células neuronais danificam-se quando a sua temperatura sobe repentinamente mais de 1 ⁰C. Esta dissertação propõem a inclusão de um sensor de temperatura em filme fino nos optrodes de μLEDs. O sensor de temperatura permitirá monitorizar a temperatura do tecido cerebral. O sensor de temperatura em filmes finos desenvolvido tem em conta as dimensões e a compatibilidade das tecnologias e materiais com os utilizados no optrode em desenvolvimento no projeto de investigação “Brain-Lighting”. O projeto “Brain-Lighting” é liderado pela empresa INOVA+ em parceria com a Universidade do Minho e a Fundação Champalimaud. Existem vários tipos de sensor de temperatura e após estudo prévio, foi escolhida a termorresistência (RTD). Foram fabricados sobre um substrato de silício sensores em dois materiais: níquel e platina. Os sensores em níquel têm dimensões na ordem dos milímetros e foram padronizados recorrendo a máscaras de sombra. Os sensores em platina foram padronizados através de técnicas de litografia, permitindo atingir as dimensões exigidas pela sonda neuronal alvo. Para os contactos dos dois tipos de sensores foi utilizado o alumínio. Os filmes-finos de alumínio e níquel foram depositados por feixe de eletrões e os filmes finos de platina foram depositados por pulverização catódica de corrente contínua. A caracterização dos sensores desenvolvidos foi realizada num forno com temperatura controlada. O método de Van der Pauw foi utilizado para determinar a resistividade dos materiais e a espectroscopia de impedância eletroquímica para determinar a impedância.
The use of MEMS technology in the manufacture of probes capable of interacting with the nervous system at the cellular level through optical stimulation is already a reality. However, these microsystems have limitations at different levels. Neural probes with optical stimulation (called "optrodes") through μLEDs allow solving the problem of freedom of movement of the subject being studied, since they allow the coupling of a wireless communication system. The problem this dissertation aims to solve is the control of the collateral heating of the neuronal tissue due to the use of μLEDs in optrodes for deep brain stimulation. Neuronal cells are damaged when their temperature suddenly rises more than 1 °C. This dissertation proposes the inclusion of a thin film temperature sensor in optrodes with μLEDs. The temperature sensor will monitor the temperature of the brain tissue. The thin film temperature sensor developed takes into account the dimensions and compatibility of technologies and materials used in the optrode under development in the “Brain-Lighting” research project. The “Brain-Lighting” project is led by INOVA + in partnership with the University of Minho and the Champalimaud Foundation. There are several types of temperature sensors and after a previous study, a resistance temperature detector (RTD) was chosen. Sensors were fabricated on a silicon substrate in two materials: nickel and platinum. The sensors in nickel have dimensions in the order of millimeters and were patterned using shadow masks. The platinum sensors were patterned by lithographic techniques, allowing to reach the dimensions required by the targeted neural probe. Aluminum was used for the contacts of the two types of sensors. The aluminum and nickel thin films were deposited by electron beam and the platinum thin films were deposited by direct current sputtering. The characterization of the developed sensors was carried out in a temperature controlled oven. The Van der Pauw method was used to determine the material resistivity and electrochemical impedance spectroscopy to determine the impedance.
