Resumo(s)O cloridrato de minociclina é um dos APIs produzidos na Cipan, sendo este produzido em maior escala. Este API é sintetizado em 7 passos, sendo que no 4º passo ocorre a formação do 7-HT. Apesar de ao longo do tempo todos os passos terem sido melhorados de forma a que o produto final cumpra todos os requisitos, gerou-se inicialmente alguns problemas no passo de síntese do 7-HT, pois este poderia estar a causar variabilidade entre lotes e assim afetar a qualidade do produto final. Foi desde então que se tornou fundamental a análise, desenvolvimento e melhoria deste passo intermediário de forma a torná-lo cada vez mais eficaz. Posto isto, desenvolveu-se posteriormente um novo método analítico para monitorizar este passo de síntese, que ao longo do tempo sofreu alterações e estudos de desenvolvimento de forma a garantir a sua viabilidade e eficácia, e com isso encontraram-se as condições ideais para se proceder à sua pré-validação. Este método apresenta características que o permitem ser versátil, com uma fase móvel compatível com LC-MS, permitindo assim a identificação de picos desconhecidos. O objetivo desta dissertação foi identificar e quantificar, não só, 7-HT como também outros produtos de síntese como a sanciclina e minociclina sal. Comparativamente ao método analítico já existente e em estudo, apenas se modificou um parâmetro cromatográfico, mudando o comprimento de onda de 350 nm para 280 nm. Através desta mudança, foi possível avaliar e perspetivar um acréscimo de informação nos cromatogramas. Outra razão desta mudança, foi o uso do mesmo comprimento de onda dos métodos tradicionais, pois assim possibilitou a comparação do novo método com o de minociclina ao mesmo comprimento de onda, 280 nm. Foram efetuadas análises comparativas para sanciclina e minociclina sal com o método tradicional Mino IPC, por forma a verificar as diferenças causadas e a viabilidade da análise destas pelo novo método. Foram determinadas linearidades para 7-HT e sanciclina, assim como fator de resposta relativo. Procedeu-se também a precisão e analise comparativa com auxílio a testes estatísticos, nomeadamente teste F para análise das variâncias e teste T para diferenças entre amostras independentes. De forma a possibilitar a utilização do novo método no controlo analítico para todas estas condições, é necessário proceder à validação do mesmo, de acordo com as guidelines regulamentares.
Minocycline hydrochloride is an active pharmaceutical ingredient (API) produced in substantial quantities in Cipan, amongst other APIs. Its production consists of seven steps, being the 4th one responsible for the formulation of 7-HT. Despite the improvements applied on the minocycline hydrochloride preparation through the years, initially there were indeed a few obstacles on the synthesis of 7-HT, regarding its variability throughout batches, affecting the quality of the final product. Therefore, it became imperative to analyse, develop and enhance the 4th step, thus increasing its effectiveness. Therefore, a new analytical method was designed to supervise the aforementioned step, which evolved over time to guarantee its accuracy and efficacy, meeting the ideal conditions to progress to its validation. The features of this method make it versatile, with a mobile phase compatible with LC-MS, making it possible to identify the unknown spikes. The purpose of this dissertation is to identify and quantify 7-HT and other synthesis compounds, as in sancycline and minocycline salt. In comparison to the analytical method previously approved, only one chromatographic parameter was modified, changing the wavelength from 350 nm to 280 nm. This adjustment permitted the evaluation and detection of additional information in the chromatographers. Using the same wavelength as the traditional methods was another factor in favor of this change, facilitating the comparison of the new and previous method of minocycline determination in the same wavelength, 280 nm. Comparative analyses were performed for sancycline and minocycline salt with the traditional Mino IPC method, with the interest of verifying the consequent differences and the analysis viability of this new method. Linearities for 7-HT and sancycline along with the relative response factor were determined. Precision and comparative analysis were also executed through statistical tests, namely the F test to address the discrepancy between variances and the T test for the divergences in independent samples. In the interest of enabling the appliance of the new method in the analytical control, it is necessary to proceed to its validation according to the current guidelines.
