Resumo(s): | TETRASPANINS (TETs) compose a large family of transmembrane proteins present in multicellular
eukaryotes. They are gaining more attention due to their considerable functional relevance. In animals,
TETs have been implicated in bringing into close proximity molecules involved in transduction of
extracellular stimuli into intracellular signalling pathways. Emerging data on animal TET structure,
genetics and biochemistry support a role in the establishment of interaction networks, organizing
protein complexes in membrane microdomains. In plants, only TETRASPANIN1 (TET1) protein is partially
characterized as having an essential role during cell differentiation and organ formation in meristems.
In this thesis, a detailed characterization of another member of the family - TETRASPANIN8 (TET8), is
presented. Key aspects, like structural features, tissue and cellular localization, and functional role of
TET8 are analysed. Several techniques and tools were implemented to fulfil this goal, particularly the use
of mutant lines: (i) loss-of-function, (ii) gain-of-function, (iii) transcriptional fusions with a GUS reporter
and (iv) translational fusions with fluorescent tags. Emphasis is given on the identification of proteinprotein
interactions, in order to gain insights on TET8 function and regulation. The putative interacting
partners were retrieved from an interaction screen using the split-ubiquitin assay and also from several
pairwise tests with known proteins.
All the data were complemented and supported by a genome-wide analysis (expression data, genome
distribution and gene duplication events) and evolutionary studies. The genome distribution of
Arabidopsis TET genes support the existence of segmental and tandem duplication events for some
genes. Furthermore, the phylogenetic analysis revealed a complex evolution of TETs in Arabidopsis, with
introns and exons forming several conserved structural patterns. Data mining of publically available
expression profiles revealed that Arabidopsis TETs, as their animal counterparts, are probably involved in
programmed cell death and plant stress response. The TET gene members have a remarkably diverse
and distinct expression pattern. It remains to be shown if they are functionally redundant or not. The diverse nature of TET8 putative interacting partners points to diverse functions. It also suggests that
TET8 might function as an adaptor protein, organizing the relative positions of partners in protein
complexes through lateral interactions in plasma membrane (PM) or in the endomembrane
compartments.
A fraction of TET8 protein is distributed in the PM, as well as in the endoplasmic reticulum (ER), Golgi and
intracellular vesicles. These vesicles reveal a dynamic movement and may correspond to the trans-Golgi
network (TGN)/endosome or multivesicular bodies (MVBs). Furthermore, these vesicles seem to be
transported along the cytoplasmatic strands in a stop-and-go movement.
The presence of TET8 in vesicles is supported by the identified interacting partners: VTI12, RAB8B, SNARE
ASSOCIATED GOLGI PROTEIN, PRA1D and AP-4μ. These proteins are involved in the regulation of protein
trafficking. The interaction with AP-4μ connects the movement of TET8-positive vesicles with the clathrindependent
pathways, present at PM and the TGN. In addition, TET8 contains a recognizable tyrosinebased
sorting motif that may be important in the recognition of TET8 as a cargo protein. The subcellular
localization pattern of TET8 makes this protein a potential candidate to establish the link between the
endocytic and secretory pathways in plant cells. Taken together, these results support the idea that TET8
may function as an accessory modulator in trafficking of the associated proteins, transducing signals
from one compartment to another.
One working hypothesis is that TET8 may be involved in plant reproduction, in particular in pollen
adhesion (pollen-stigma recognition), pollen tube formation and/or fertilization. The absence of tet8
homozygous mutant lines indicates that the mutation is probably lethal and can only be propagated in
heterozygous individuals. Moreover, the mutation is not affecting the development of the female ovules.
The working hypothesis is supported by the reported involvement of secretory and endocytic pathways
in pollen recognition, pollen tube growth and fertilization. Knockout mutations in genes that encode
proteins involved in endomembrane trafficking frequently show no obvious mutant phenotype under
normal conditions, but are often embryonic or gametophytic lethal.
The common origin and structure of animal and plant TETs, indicates that they should share a
molecular function. In this study, this assumption is partially validated, since TET8 has putative
interacting partners similar to others reported for animal TETs, in particular for the CD63 member.
The complementary approaches used in this work to investigate TET8 function has broaden our current
knowledge about TET biology in plants. However, the key features of plant TETs have just begun to be
uncovered and raised many new questions for further research. As TETRASPANINAS (TETs) compõem uma família de proteínas transmembranares, com relevância funcional e que se encontram presentes em organismos eucariontes multicelulares. Nos animais, as TETs têm sido implicadas no estabelecimento de interacções que são responsáveis pela aproximação de moléculas envolvidas na transdução de estímulos extracelulares com outras das vias de sinalização intracelular. Descobertas recentes sobre a estrutura, genética e bioquímica das TETs de animais indicam que o seu papel consiste na promoção de uma densa rede de interacções, organizando complexos proteicos presentes em microdomínios membranares. Nas plantas, apenas a TETRASPANINA1 (TET1) está parcialmente caracterizada, sendo uma proteína essencial na diferenciação celular e formação de órgãos ao nível dos meristemas. Nesta tese, é apresentada uma caracterização detalhada de um outro membro desta família – TETRASPANINA8 (TET8). Os aspectos essenciais, como as propriedades estruturais, a localização tecidular e celular e o seu papel funcional são analisados. Para cumprir este objectivo, diversas técnicas e procedimentos foram implementados, em particular o uso de linhas mutantes: (i) loss-of-function, (ii) gain-of-function, (iii) fusões com o repórter GUS e (iv) fusões com proteínas repórteres fluorescentes. Especial ênfase foi colocada na identificação de interacções proteína-proteína, de forma a obter informação sobre a função e regulação da proteína TET8. Os presumíveis parceiros de interacção foram detectados a partir de um rastreio de interacções ou testando proteínas conhecidas usando o sistema split-ubiquitin. Todos os dados obtidos foram complementados e suportados por uma análise genómica (dados de expressão génica, distribuição no genoma e eventos de duplicação génica) e estudos de evolução em Arabidopsis. A distribuição genómica dos genes desta família suporta a ocorrência de duplicações para alguns dos genes. Por sua vez, a análise filogenética revelou uma evolução complexa das TETs em Arabidopsis, com os intrões e exões a evidenciarem um padrão estrutural conservado. A análise de perfis de expressão génica disponíveis em bases de dados públicas revelou que as TETs de Arabidopsis, tal como as de animais, estão provavelmente implicadas na morte celular programada e na resposta da planta a diferentes stresses. Os membros da família possuem um padrão de expressão muito diverso e distinto, necessitando ainda de ser comprovado se serão funcionalmente redundantes ou não. A natureza diversa dos presumíveis parceiros de interacção indica uma miríade de funções. Também sugerem que a proteína TET8 poderá funcionar como um adaptador, organizando a posição relativa dos parceiros nos complexos proteicos, através de interacções laterais na membrana plasmática ou nos compartimentos intracelulares. Uma fracção da proteína TET8 encontra-se na membrana plasmática, no retículo endoplasmático, no complexo de Golgi e em vesículas do sistema endomembranar. Estas vesículas revelam um movimento dinâmico e podem corresponder ao compartimento trans-Golgi (TGN)/endossomas ou aos corpos multivesiculares (MVBs). Estas vesículas são, aparentemente, transportadas pelos fluxos de citoplasma, num movimento não contínuo e que evidencia paragens (stop-and-go). A presença da proteína TET8 em vesículas é igualmente apoiada pelos parceiros de interacção detectados: VTI12, RAB8B, SNARE ASSOCIATED GOLGI PROTEIN, PRA1D e AP-4μ. Estas proteínas encontram-se associadas à regulação do transporte vesicular. A interacção com a subunidade AP-4μ estabelece a conexão entre o transporte das vesículas com as vias dependentes da clatrina, as quais se encontram activas na membrana plasmática e no trans-Golgi. Estruturalmente, a sequência da TET8 apresenta domínios que podem estar associadas ao reconhecimento da TET8 enquanto proteína cargo. O padrão de localização intracelular da TET8 corrobora o seu possível envolvimento no estabelecimento de ligações e regulação das vias de secreção proteica e de endocitose nas células vegetais. Analisando de forma global, os resultados apoiam a ideia da TET8 ser uma proteína que modela o transporte de proteínas associadas, podendo ser importante na comunicação ao nível celular, transmitindo sinais de um compartimento para outro. A hipótese de trabalho implica a TET8 na reprodução vegetal, em particular na adesão do pólen (reconhecimento entre o pólen e o estigma), formação do tubo polínico e/ou fecundação. A ausência de linhas homozigóticas tet8 levou a postular que a mutação do gene TET8 é letal e propagada apenas em indivíduos heterozigóticos. A mutação não afecta o desenvolvimento dos gâmetas femininos. A hipótese de trabalho é suportada pela actividade intensa das vias secretória e endocítica que são amplamente conhecidas no desenvolvimento dos processos anteriormente designados. São conhecidos diversos genes associados ao transporte endomembranar que, quando são silenciados, não originam fenótipos mutantes em condições normais de desenvolvimento, mas que se revelam letais durante o desenvolvimento dos gâmetas ou do próprio embrião. A origem comum das TETs animais e vegetais, associadas a uma estrutura semelhante, leva a postular que estas proteínas devem partilhar a mesma função molecular. Nesta tese, esta hipótese é parcialmente validada, pois a proteína TET8 possui parceiros de interacção semelhantes a outras TETs de animais, com destaque para a CD63 (TET humana). As estratégias complementares usadas para investigar a função da proteína TET8 expandiram o nosso conhecimento sobre a biologia das TETs nas plantas. No entanto, as características destas estão apenas a começar a ser reveladas, levantando muitas questões para investigações futuras. |
