Resumo(s): | No presente trabalho pretendeu-se sintetizar um dua/-Iinker contendo uma unidade fluorescente bifuncional que pudesse ser fotoclivada selectivamente a diferentes comprimentos de onda, permitindo a separação e quantificação por HPLC com detecção UV/Vis ou fluorescência. Testaram-se duas cumarinas, nomeadamente a 1-hidroximetil-6-hidroxicumarina e a 1-hidroximetil-6-metoxicumarina, que são conhecidas pela sua gama espectral alargada, rendimentos quânticos de fluorescência elevados e fotoestabilidade. Foi investigada a sua aplicação a uma resina fotoclivável comercial e na ligação a aminoácidos modelo, nomeadamente a Lfenilalanina e a L-valina na cadeia principal, e a L-serina na cadeia lateral, através das funções amina, ácido carboxílico e hidroxilo dos aminoácidos.
Foi investigado o comportamento destes derivados fluorescentes face a diferentes condições de clivagem, nomeadamente o comprimento de onda de irradiação e diferentes solventes orgânicos, num reactor fotoquímico equipado com lâmpadas de radiação monocromática de 254, 300 e 350 nm. Foram também efectuados estudos cinéticos de clivagem nas condições referidas por monitorização por HPLC com detecção UV. In the present work we envisaged the synthesis of a dual-linker bearing a bifunctional fluorescent moiety which could be selectively photoreleased at different wavelengths, allowing separation and quantization of products by HPLC with UV/Vis or fluorescence detection. Two coumarins were tested, namely 1-hydroxymethyl-6- hydroxycoumarin and 1-hydroxymethyl-6-methoxycoumarin, which are known for their extended spectral range, high emission quantum yields and photostability. Its application to a commercially available photocleavable resin and in the linkage to model amino acids, namely L-phenylalanine and L-valine at the main chain, and Lserine at the side chain, through the amino, carboxylic acid and hydroxyl functions of the amino acids was investigated. The behaviour of these fluorescent derivatives was studied in different cleavage conditions, namely the wavelength of irradiation and the use of different organic solvents, in a photochemical reactor equipped with monochromatic Iamps of 254, 300 and 350 nm. Cleavage kinetics studies were undertaken in the above mentioned conditions and monitored by HPLC/ UV detection. |
