Resumo(s): | Cell-material interactions are responsible for the success of many biomedical applications. Within
the properties of the biomaterial substrates, surface topography has been reported to have paramount
importance in this interaction. Cell adhesion, cell morphology, cell proliferation, cell differentiation, and
immunogenicity are important cell functions that are reported to be affected by materials’ surface
topography. Artificially generated topographies have been studied but biomimetic topographies can bring
new insights into the mechanisms that underlie the cellular response to the topography of the substrates.
Natural surfaces present a unique ability for the development of substrates with unique hierarchical
arrangements of topographical features at different length scales that hardly can be achieved in artificially
generated topographies.
In this thesis, we applied the replica molding technique followed by the Nanoimprint Lithography to
replicate biological surface topographies on Polycaprolactone (PCL) membranes. PCL membranes with
five different biomimetic surface topographies were produced: Rubus fruticosus leaf, E coli,
S.eppidermidis, L929 cells and an Eggshell membrane (ESM). PCL membranes imprinted Rubus
fruticosus leaf surface topography revealed to be cytocompatible and to have a higher angiogenic capacity
when compared with flat surfaces. Moreover, those membranes were able to drive osteogenesis of
mesenchymal stem cells without any biochemical stimulation, presenting a synergistic effect with the
osteogenic induction medium. The surface topography of E. coli, S.eppidermidis and L929 cells and an
ESM, all of them imprinted in PCL membranes, were studied to evaluate its potential for the modulation
of macrophages' inflammatory response. Our results revealed that depending on the surface where these
cells are seeded, they present different phenotypes. Porous PCL membranes imprinted with the surface
topography of the ESM were also produced intending to generate a biomimetic analog of basement
membranes. The generated membranes, besides the selective molecular weight permeability, revealed
being able to support cell proliferation. Their surface topography also induces the production of relevant
extracellular matrix proteins.
Altogether, the works developed in this thesis enabled the generation of cell culture substrates with
different biomimetic surface topographies that showed to have relevant impacts on biological performance
opening new avenues to tailor high-performance substrates for biomedical applications. As interações entre células e biomateriais são responsáveis pelo sucesso da sua aplicação na área biomédica. A topografia de superfície tem sido apontada com sendo crucial neste tipo de interações. Adesão, morfologia, proliferação e diferenciação celular, assim como a imunogenicidade são importantes funções celulares que se evidenciam como sendo afetadas pela topografia de superfície dos materiais. Ao contrário das topografias criadas artificialmente, as topografias biomiméticas replicam estruturas naturais, pelo que podem trazer novos conhecimentos sobre mecanismos específicos que estão subjacentes à resposta celular associadas a essas estruturas. Além disso, topografias naturais apresentam topografias únicas com arquiteturas hierárquicas com diferentes tamanhos e escalas relevantes para influenciar processos biológicos e celulares. Nesta tese, utilizámos o método de replicação por molde seguido de litografia por nanoimpressão para replicar a topografia de superfícies naturais em membranas de policaprolactona (PCL). Foram produzidas cinco tipos de membranas de PCL com topografias de superfície biomiméticas: da folha de Rubus fruticosus, de E. coli, de S. eppidermidis, de células L929 e da membrana da casca do ovo (ESM). Membranas de PCL impressas com a topografia de superfície da folha de Rubus fruticosus revelaram ser citocompatíveis e ter maior capacidade angiogénica quando comparadas com topografias lisas. Estas membranas demonstraram também ser capazes de induzir a diferenciação osteogénica de células estaminais mesenquimais, apresentando um efeito sinergístico com o meio de indução da osteogénese. Membranas de PCL impressas com a topografia de superfície de E. coli, de S. eppidermidis, de células L929 e da ESM foram utilizadas para estudar o seu potencial modulatório na resposta inflamatória dos macrófagos. Os nossos resultados revelaram que dependendo da superfície onde estas células são cultivadas, elas apresentam diferentes fenótipos. Foram também produzidas membranas de PCL porosas impressas com a topografia de superfície da ESM com o objetivo de criar um análogo biomimético de membranas basais. Estas membranas apresentam uma permeabilidade seletiva para o peso molecular, conseguem suportar a proliferação celular e a sua topografia induz a produção de proteínas da matriz extracelular. Os trabalhos desenvolvidos nesta tese permitiram criar substratos para cultura de células com topografias de superfícies biomiméticas que demonstraram ter impacto no desempenho biológico, abrindo novos caminhos para obter substratos com alto desempenho de para aplicações biomédicas. |
