| dc.description.abstract | O metano é produzido a partir de uma ampla diversidade de resíduos orgânicos por comunidades microbianas anaeróbias, onde as relações sintróficas desempenham um papel determinante na eficiência do processo. Os mecanismos de transferência de eletrões interespécies (IET) podem envolver a troca de transportadores de eletrões (hidrogénio, formato), mas também a transferência direta de eletrões entre espécies (DIET). Os materiais condutores melhoram as taxas de produção de metano (MPR), mas os mecanismos envolvidos nessa IET mediada pelos materiais ainda são controversos e não totalmente conhecidos. Nesta tese, investigou-se o efeito de vários (nano)materiais, nomeadamente carvão ativado (AC), magnetite (Mag), nanotubos de carbono (CNT), AC/CNT modificados, zeólito (Zeo), areia e esferas de vidro, na produção de metano (MP). Apesar das diferenças a nível físico-químico, em geral, os (nano)materiais aumentaram as atividades metanogénicas, diminuindo as fases de latência que antecedem a MP e aumentando as MPR. As diversas características físico-químicas dos (nano)materiais, como a área de superfície específica, a área dos mesoporos, o volume dos microporos, a condutividade elétrica, a acidez/basicidade do material e as impurezas presentes na sua composição (por exemplo, S e N), isoladamente não conseguem explicar as melhorias observadas na MP. O silício foi um elemento encontrado em todos os materiais testados e melhorou a MP por mecanismos ainda desconhecidos, que merecem ser investigados. As mudanças observadas no potencial redox do meio de crescimento na presença de (nano)materiais não se correlacionaram diretamente com a melhoria na MP. Além disso, diferentes (nano)materiais podem afetar de forma diferente o mesmo microrganismo metanogénico. Por exemplo, o Methanobacterium formicicum foi bastante estimulado pelo AC e drasticamente inibido pela Mag. Contudo, o mesmo (nano)material não afetou outras espécies da mesma forma. Por exemplo, o Zeo melhorou a MPR do M. formicicum (a crescer em H2/CO2) e da Methanothrix harundinacea (a crescer em acetato), mas não teve efeito sobre a MP do Methanospirillum hungatei (a crescer em H2/CO2) e da M. barkeri (a crescer em acetato). Os resultados sugerem ainda que as espécies microbianas são afetadas diferencialmente, independentemente do tipo de substrato. O efeito dos (nano)materiais também varia dependendo da cocultura sintrófica, uma vez que o AC aumentou a atividade metanogénica da cocultura sintrófica Syntrophomonas zehnderi e M. formicicum, mas a Mag causou uma drástica inibição. Já noutra cocultura, constituída por Syntrophomonas wolfei e M. hungatei, o AC e a Mag não causaram impacto significativo. A atividade microbiana também parece influenciar o efeito dos (nano)materiais, já que melhorias significativas foram observadas principalmente por culturas menos ativas. Por exemplo, o AC melhora as MPR do M. formicicum quando adicionado na altura da inoculação, mas não teve efeito quando adicionado a culturas ativas deste microrganismo durante a fase de crescimento exponencial. Esta relação entre a atividade microbiana e o efeito dos (nano)materiais também foi observada em enriquecimentos microbianos e no bioreator operado em contínuo. A composição microbiana dos enriquecimentos especializados na degradação de butirato e desenvolvidos com e sem (nano)materiais foi diferente, e não foram enriquecidas espécies eletroativas (tais como Geobacter spp.) na presença dos materiais. | dc.description.abstract | Methane is produced from a wide diversity of organic wastes by anaerobic microbial communities, where syntrophic relationships play a key role in the efficiency of the process. Interspecies electron transfer (IET) mechanisms may involve the exchange of electron carriers (hydrogen, formate), but also direct interspecies electron transfer (DIET). Conductive materials improve methane production rates (MPR), but the mechanisms involved in this material mediated IET are still controversial and not fully known. In this thesis, the effect of several (nano)materials, namely activated carbon (AC), magnetite (Mag), carbon nanotubes (CNT), modified AC/CNT, zeolite (Zeo), sand and glass beads, on methane production (MP) was investigated. Despite their physicochemical differences, generally, (nano)materials enhanced methanogenic activities, by decreasing lag phases preceding MP and increasing MPR. The various physicochemical characteristics of (nano)materials, such as the specific surface area, area of mesopores, volume of micropores, electrical conductivity, material acidity/basicity, and impurities present on material composition (e.g., S and N) could not explain per se the enhancement observed on MP. Silicon was an element found in all materials tested and, per se, improved MP by mechanisms yet unknown, which deserve to be further explored. The changes observed on redox potential of the growth medium in the presence of (nano)materials were not directly correlated with the improvement of MP. Besides, different (nano)materials can impact differently the same methanogen. For example, Methanobacterium formicicum was greatly stimulated by AC and drastically inhibited by Mag. However, the same (nano)material did not impact all species in the same way. For instance, Zeo improved MPR by M. formicicum (growing on H2/CO2) and Methanothrix harundinacea (growing on acetate) but had no effect on MP by Methanospirillum hungatei (growing on H2/CO2) and M. barkeri (growing on acetate). These results likewise suggest that microbial species are differentially affected, independently of the type of substrate. The effect of (nano)materials also varies depending on the syntrophic coculture, since AC enhanced the methanogenic activity of the syntrophic coculture Syntrophomonas zehnderi and M. formicicum, but Mag caused severe inhibition. Yet, in another coculture, composed by Syntrophomonas wolfei with M. hungatei, AC and Mag did not cause significant impact. The microbial activity also seems to dictate the effect of (nano)materials, as significant improvements were mainly observed by less active cultures. For instance, AC generally improved MPR of M. formicicum cultures, but had no effect when added to highly active M. formicicum cultures during the exponential growth phase. This relation between microbial activity and the effect of (nano)materials was also observed in enrichment cultures and in the continuous bioreactor. Microbial composition of butyrate-degrading enrichments developed with and without (nano)materials was different, but electroactive species (e.g., Geobacter) were not enriched in the presence of any material. Microscopic, taxonomic and gene expression analyses suggested that butyrate conversion to hydrogen and acetate occurred by microorganisms closed related to Syntrophomonas, and no genes related to DIET were detected. | por |
