| dc.description.abstract | Pancreatic ductal adenocarcinoma (PDAC) is the most common and deadly form of pancreatic cancer with a survival rate of approximately 5%, being one of the most resistant cancers to conventional treatment. Metal-based compounds have attracted attention as potential anticancer agents. PMC79, a ruthenium-based compound developed by our group, has demonstrated selective anticancer effects in KRAS-mutated colorectal and pancreatic cancer cell lines. Given the high resistance of PDAC to single-agent treatments, combination therapy represents a promising strategy to increase therapeutic efficacy. The aim of this work was to assess the effects of PMC79 in combination with gemcitabine, exploring if the combined treatment exhibits increased potential anticancer effect in KRAS-mutated PDAC cells and evaluate the effect of PMC79 in KRAS and its downstream molecules. Combined treatment with gemcitabine and PMC79 significantly reduced cell viability, particularly in KRAS-mutated PDAC cell lines. This combination also impaired significantly colony formation ability, decreasing both the number and size of colonies compared to individual treatments. Cell cycle analysis revealed distinct responses among cell lines, with the combination of gemcitabine and PMC79 inducing an S phase cell cycle arrest in MIA PaCa-2 (KRASG12C) and PANC-1 (KRASG12D), reflecting the synergistic effect of the two compounds s in these cell lines. The combination of gemcitabine and PMC79 induced apoptosis in KRAS-mutant PDAC cell lines, particularly in MIA PaCa-2 (KRASG12C) and Capan-2 (KRASG12D) Our results on the analysis of the effect of PMC79 on KRAS and KRAS signalling molecules suggest that PMC79 exerts selective cytotoxic effects by inhibiting KRAS and modulating downstream signalling pathways, PI3K/AKT and MAPK/ERK, in a time and dose-dependent manner in all PDAC cell lines Overall, these results suggest that the combination of PMC79 and gemcitabine represents a promising therapeutic strategy for KRAS-mutated PDAC, potentially allowing the use of lower gemcitabine doses and thereby reducing associated adverse effects. This approach could address a significant clinical challenge and offer a novel treatment option for patients with PDAC harbouring KRAS mutations. | eng |
