Aerobic granular sludge (AGS) is a novel compact technology presenting sustainability potential over conventional aerobic processes. However, a long start-up for granules formation and their long-term stability are still the main issues for a wider establishment of this technology. AGS characterization and evaluation have gained attention to improve our understanding and process control. In this thesis, quantitative image analysis (QIA) methods were used to define and quantify the morphological changes of sludge during granulation processes and evaluate the stability of granules under specified operating conditions. For this purpose, granulation was initially monitored in two sequencing batch reactors (SBR) at different organic loading rates (OLR) and minimal imposed sludge settling velocities (MISSV). While the stratification patterns and the morphology of the initial small granules revealed the turning point from flocculation to granulation, multivariate statistical analysis of the QIA data revealed the sludge aggregation and granules maturation status and anticipated the sludge settling properties. Moreover, a complex relationship between nutrient removal and sludge morphology was assessed during granulation under different chemical oxygen demand (COD)-to-nitrogen ratios (COD/N) and washout dynamics. Poor nitrogen removal efficiencies during the formation of aerobic granules favoured the development of viscous and filamentous bulking, which impaired the granulation process at the early stages. The stability and activity of mature granules as bioaugmenting microbial consortia (i.e., bioactive granules) were evaluated in AGS systems exhibiting poor nutrient removal capabilities. The morphology of the bioactive granules and innate AGS were fundamental for bioaugmentation success in AGS systems. The stability of AGS is a bottleneck of AGS systems. Thus, the long-term stability of mature granules was evaluated in the presence of increasing oleate concentration. QIA data revealed that oleate adsorption onto AGS influenced granules distribution patterns, inducing selective sludge washout of small granules owing to their larger specific surface area. Moreover, principal component analysis (PCA) revealed the morphological changes of the AGS due to oleate, which were correlated with decreasing reactor performance. Overall, QIA increased our understanding of the sludge morphological transition during the granulation process, as well as their stability under defined operating conditions, highlighting the potential of QIA to monitor and regulate AGS systems.por
