| dc.description.abstract | Nanomaterials have propelled deep changes in the technological, industrial and medical paradigm, which was accompanied by its translation from simple structural substituents to versatile multifunctional materials. Currently, nanomaterials are sought as solutions to modern medical problems through the convergence of materials science with biology. Supramolecular peptide-based hydrogels are stimulus-responsive self-assembled intertwined fibrillar structures achieved through the cooperative effect of different non-covalent intermolecular interactions. These are among the most important classes of biomaterials owing to the wide range of applications, which include, among others, drug delivery, tissue engineering, in vivo imaging and template materials. The combination with magnetic nanoparticles affords magnetogels, which ensure efficient transport and accumulation of drugs in the therapeutic sites or localization by using an external magnetic field gradient, thus enhancing the specificity of anticancer drugs, most of them toxic and with systemic side effects. Moreover, the addition of a gold shell to the magnetic nanoparticles improves biocompatibility and allows photothermia therapy, which can be used as a photo-triggered release mechanism of the encapsulated drug. In this work, core/shell manganese ferrite/gold and gold-decorated manganese ferrite nanoparticles were developed, and their structural, spectroscopic and magnetic properties were characterized. The nanoparticles were combined with two new self-assembled peptide-based hydrogels. The hydrogelators were spectroscopically characterized and evaluated as anti-inflammatory agents by computational tecniques. The hydrogels and magnetogels were structurally characterized and tested as nanocarriers for curcumin (an anticancer and neuroprotective potential drug). The systems that were suitable carriers were also screened as potential multimodal therapeutic agents. Screening assays included the interaction with biomembrane models, determination of magnetic and rheological properties, drug release assays and photothermia ability. The obtained results are promising for the use of the new magnetogels in multimodal cancer therapy, owing to the high-loading capacity, synergistic magnetically-guided drug delivery, magnetic hyperthermia and photothermia, which are expected to improve therapeutic efficiency in cancer therapy. | por |
